In order to do this you have to be in the Create Text Tool located in the Toolbox Bar (Bar along the left side of the Inkscape Interface), it is represented by the Letter A icon.
Create your text, stay in this tool and DRAG your mouse over the character you want to change the color of. Open the Fill & Stroke Menu (Shift+Ctrl+F). Now change the single characters fill color.. done..

In 0.47 it will have created a group of paths, one for each letter. Double-click to enter the group, select the letter you want to change, then use the Fill and Stroke dialogue or the colour palette as with any other object.
Prior to 0.47 text converted to a path resulted in a single path object. You can use "Path=>Break Apart" to split it into separate paths for each letter, then select and modify the one you want to. Be aware, however, that this approach can cause problems with letter that have a "hole" in them. If you experience that issue, don't panic. The hole still exists, but now it's a separate path. Select both it and the main body of the letter and use "Path=>Difference" to cut the hole out again.
