hi,
i have been working on this image and want to add 3d effect and i would like to have more realistic(natural looks) design.
1.i want the outer tube to be look like a sealed glass tube
2.i want that green liquid to be fluorescent green liquid inside the glass tube
3.i want the text to be half sink in the liquid.

Oh, well I think the green already does look like liquid, and may not need much tweaking, once the rest of the image is like you want.
If it is a sealed glass tube, I don't understand the gray shading on top. I think instead, you might want white or white-ish highlights, and maybe even some reflections. You might look around for some tutorials on making things appear like glass (especially try those in my signature).
The part of the image that's white, inside the tube, above the green -- should that appear transparent, so that you would see the color of whatever background you have, seen through the tube there? (Perhaps it took on the white when you saved as PNG in order to upload it? If so, you should use File menu > Export Bitmap instead.)
I think you've made a great start, and it will be one step at a time. Plus I'm sure others will have some helpful comments as well. I'll be interested to see the final product!

Well, I have no idea if there is a tutorial for drawing a glass tube. What I meant was that you might find some tips for making things look like glass, by looking at tutorials for making glass-looking things. For example, I've seen a tutorial somewhere for making a glass orb.

That is a challenging thing to draw, but a fun challenge.
You might look at test tubes for a visual reference.
Here is a test tube image (that I did not draw and might be copyrighted although I think it is a wallpaper) that might help.
Also look-up "wave machine", have your ever seen those desktop wave machines, they have a really 3d wave.
In black and white photography they used to say you need a true black and a true white in your photo.
Just the same, you might need to make sure your drawing spans the full tonal range to really hit that 3d effect.
